A Whey acidic protein promoter (Mouse)/Insulin-like growth factor 1 (Rat) Transgenic Mutation


Summary
Insulin like growth factor-1 (IGF1) is a mitogen for several breast cancer cell lines, and IGF1 receptors have been found on primary human breast cancer specimens. It has been suggested that estrogen-independent breast cancer cell proliferation is mediated by interactions between several growth factors, including IGF1, TGFA and TGFB. A family of IGF binding proteins modulate IGF1 biological activity by sequestering or transporting the growth factor. IGFBP3 is a major carrier of IGF1 and potentiates its actions.

Background


Transgene
The targeting constuct consister of a rat prepro Igf1 cDNA ligated to a 7 kb EcoR1 fragment of the mouse Wap gene. The construct was microinjected into the pronuclei of zygotes from (C57BL/6 x SJL)F1 females.

Mammary Phenotype

The transgene is expressed at high levels during lactation and post-lactation. Homozygotes are viable and fertile. There were no obvious changes in mammary gland histomorphology during lactation, but at 5 to 10 days post-laction, there was a delay in involution and decreased apoptosis.


Controls
The genetic background of this strain is mixed C57BL/6 x SJL. Normal wildtype siblings or B6SJLF1/J mice may be used as controls. The B6SJLF1/J mice only provide an approximate genetic match to this B6,SJL background.
